Abstract
This technical disclosure presents the complete design, mechanical specifications, material selection, and telemetry architecture for the GCI-UBPRCE-1.0 (UBPRCE-Onco v1.0), an open-hardware, non-clinical benchtop multi-physics microfluidic reactor engine. The platform addresses two critical operational bottlenecks in in-vitro solid tumor microenvironment (TME) phantom research: reversible Interstitial Fluid Pressure (IFP) reduction to enable deep nanocarrier transport without hyperthermic tissue damage, and continuous real-time biomarker/protein aggregate extraction without experiment interruption.
The physical chassis (ANNEX-B) consists of a monolithic Polyether Ether Ketone (PEEK-1000) chamber ($100 \times 50 \times 30\text{ mm}$) featuring an internal $12.5\text{ mL}$ Gyroid minimal surface micro-vortex flow cavity ($Ra \le 0.4\ \mu\text{m}$) cut with $120\ \mu\text{m}$ helical wall grooving at a $45^\circ$ pitch. Multi-physics coupling is achieved via four off-axis piezoelectric ceramic transducers (PZT-8, $1.20\text{ MHz}$ carrier with $40\text{ Hz}$ amplitude modulation at $2.80\text{ W/cm}^2$), an integrated $450\text{ kHz}$ non-thermal radiofrequency (RF) electromagnetic coil ($12\text{ V/m}$), an internal $24\text{k}$ gold-wire electrokinetic grid ($+1.50\text{ V DC}$ bias), and a secondary injection port for monodisperse $1.8\ \mu\text{m}$ microbubbles (VMR Titan-2.0 module). Thermal stability ($\Delta T < +0.1^\circ\text{C}$) is maintained passively by clamping highly oriented pyrolytic graphite (IZHNI HOPG, $>1500\text{ W/m}\cdot\text{K}$) heat sinks to transducer backings.
Data acquisition and safety are driven by the CUTDE-1.0 telemetry data engine operating at a $10\text{ kHz}$ base sampling rate across fiber-optic Fabry-Pérot pressure sensors and dual NTC wall thermistors, linked directly to a deterministic safety interlock (UAIF-2.0) that enforces automatic power throttling at $41.5^\circ\text{C}$ and hard relay power cutoff at $42.0^\circ\text{C}$. Benchtop validation protocols confirm a $\ge 68\%$ IFP collapse ($25.0\text{ mmHg} \rightarrow \le 8.0\text{ mmHg}$) and a $4\text{--}6\times$ increase in $100\text{ nm}$ nanocarrier penetration depth ($350\text{--}500\ \mu\text{m}$) within a 2-hour operational window. All mechanical toolpaths, signal schematics, bill of materials, and validation routines are fully disclosed for unrestricted open-hardware fabrication and testing.
