Abstract

2-n-Butyl-3-[4-(3-di-n-butylaminopropoxy)benzoyl]-5-aminobenzofuran dioxalate is a key intermediate in the synthesis of dronedarone hydrochloride. Process development studies were undertaken to evaluate the hydrogenation of 2-n-butyl-3-[4-(3-di-n-butylaminopropoxy)benzoyl]-5-nitrobenzofuran under various catalytic conditions. The influence of catalyst loading, hydrogen pressure, temperature, reaction medium, and acidic additives was assessed. Successful conversion of the nitro intermediate to the corresponding amino derivative was observed under a range of operating conditions, demonstrating the flexibility and robustness of the hydrogenation process.
