Abstract
VORTEX-AC is a field-deployable, single-cartridge diagnostic platform for Yersinia pestis (pneumonic, bubonic, and septicemic plague) that integrates passive clog-bypass geometry, Dean-vortex micro-mixing, tilted-angle surface acoustic wave (taSSAW) concentration, vortex-enhanced lysis, and loop-mediated isothermal amplification (LAMP) into a single PDMS cartridge on a TPMS-structured thermal base with a phase-change material (PCM) buffer.
The system targets a 33-minute total time-to-result from raw sputum load to naked-eye colorimetric readout, with a projected 10–100× improvement in limit of detection (LOD) over current LAMP assays due to acoustic concentration of target bacteria prior to amplification.
The novel integration — acoustic concentration upstream of isothermal amplification for Y. pestis — has not been published. No existing plague test (F1RDT, LAMP, qPCR, or FA-RMP platform) uses acoustic concentration to boost effective target concentration before amplification. The TPMS Gyroid thermal spreader + PCM buffer combination for LAMP is also unpublished.
