Abstract

The Vortex Reprogramming Platform (VRP-1.0) is an open-hardware, closed-loop research system designed to deliver p53-preserving chemical reprogramming agents to aged human cells using acoustic-vortex microfluidic forces, while simultaneously monitoring cell state in real time and clearing the lipid droplet accumulation that published research identifies as a primary barrier to successful in vivo chemical rejuvenation.

The platform integrates five novel elements not found in any currently published reprogramming system:

  1. Pre-treatment oncogenic pre-screening gate — removes high-risk cells before reprogramming begins, not after (proactive, not reactive safety)

  2. Acoustic-vortex intracellular delivery of chemical reprogramming agents without viral vectors, following the AESOP physics (UC Irvine, Lab on a Chip 2026)

  3. Real-time p53/p21 activity monitoring via acoustic stiffness sensing — distinguishing the rejuvenation zone from the senescence zone during treatment

  4. Post-reprogramming lipid droplet clearance stage — directly addressing the toxic lipid accumulation identified as a rejuvenation barrier (bioRxiv 2025)

  5. Nuclear compliance tracking as a positive biological endpoint — giving the controller something to aim for, not just avoid

All hardware is built from open, commercially sourceable, or fabricatable components. No viral vectors. No gene editing. No proprietary reagents beyond the chemical reprogramming cocktail, which is fully cited.

After reading this document, a qualified cell biology / biomedical engineering lab team can build the Phase 1 benchtop and run the validation protocol.
